Drosophila hematopoietic progenitor is maintained both by niche-derived neighboring factors and by systemic interactions. Previous study have identified that olfactory receptor neurons (ORN) functions upstream of a small set of neurosecretory cells that express GABA. Upon olfactory stimulation, GABA from these neurosecretory cells is secreted into the hemolymph and binds to metabotropic GABAB receptors expressed on blood progenitors within the lymph gland and functions upstream of cytosolic calcium. Loss of olfactory signal reduces GABA secretion from the brain and cytosolic calcium concentration, in turn causes precocious differentiation of blood progenitors. On the contrary, constitutive active ORN (Olfactory Receptor Neuron) produces excess GABA, and elevated GABA induces differentiation of progenitors mediated by accumulation of high ROS. These studies indicate that moderately high level of GABA is required for the blood progenitor maintenance, and establishe signaling roles for GABA in the regulation of blood progenitors. Given the presence of GABA in mammalian blood circulation, our study leads to a model that could be extended to understand different roles of non-neuronal GABA in mammalian development and stress response.
